Is it possible to have the following?:
Folder setup, with SFTP access ONLY for a specific user
Same folder setup with FTP access (SFTP being an option is fine for this as well) for another user?

If you want one user to only have SSH/SFTP and not FTP (and vice versa for the other user) you would need to setup two separate domains.
If you don't mind them accessing both SSH/SFTP and FTP at the same time you can do it within the same domain.
